However, this is not something new. The origin of vaccines dates back to 1802 when <strong>Edward Jenner revolutionized the fight against smallpox<\/strong><span style=\"font-weight: 400\">. He observed that people who worked with cattle often did not contract the disease during smallpox outbreaks since when dealing with cattle infected with <\/span><strong>cowpox<\/strong><span style=\"font-weight: 400\">, they developed <\/span><strong>mild pustules<\/strong><span style=\"font-weight: 400\"> in the body which in turn <\/span><strong>immunized<\/strong><span style=\"font-weight: 400\"> them <\/span><strong>against human smallpox<\/strong> (much more virulent).<\/p><\/p>\n\n\n\n<p><strong>Technological advances have made it possible to refine the methods to design vaccines<\/strong>, and today there are 3 main methods.&nbsp;<\/p>\n\n\n\n<p class=\" translation-block\"><strong>Technological advances have made it possible to refine the methods to design vaccines<\/strong>, and today there are 3 main methods.<\/p>\n\n\n\n<p>Before going into explaining how each vaccine works, it is important to know the basic central dogma of molecular biology.<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">Central Dogma of Molecular Biology<\/h2>\n\n\n\n<figure class=\"wp-block-image size-large\"><img fetchpriority=\"high\" decoding=\"async\" width=\"1024\" height=\"419\" src=\"https:\/\/adntro.com\/wp-content\/uploads\/2022\/04\/dogma-central-Biologia-molecular-1024x419.png\" alt=\"Central Dogma of Molecular Biology\" class=\"wp-image-6251\" srcset=\"https:\/\/adntro.com\/wp-content\/uploads\/2022\/04\/dogma-central-Biologia-molecular-1024x419.png 1024w, https:\/\/adntro.com\/wp-content\/uploads\/2022\/04\/dogma-central-Biologia-molecular-300x123.png 300w, https:\/\/adntro.com\/wp-content\/uploads\/2022\/04\/dogma-central-Biologia-molecular-768x315.png 768w, https:\/\/adntro.com\/wp-content\/uploads\/2022\/04\/dogma-central-Biologia-molecular-18x7.png 18w, https:\/\/adntro.com\/wp-content\/uploads\/2022\/04\/dogma-central-Biologia-molecular.png 1299w\" sizes=\"(max-width: 1024px) 100vw, 1024px\" \/><figcaption class=\"wp-element-caption\">In humans, DNA constitutes our genetic material, but there are certain viruses such as SARS-Cov-2 whose genetic code is stored in the form of a <a href=\"https:\/\/adntro.com\/en\/blog\/genetic-curiosities\/arn-types\/\">RNA<\/a>. Both DNA and RNA constitute the genetic material and are transcribed to form a molecule known as messenger RNA (mRNA). This molecule is transient and serves to originate little by little all the resources necessary for the human being or virus to live. Each mRNA has instructions to originate a specific element (protein) that by itself serves no purpose, but is essential to form the functional assembly that allows us to live. Therefore, we could say that both DNA and mRNA or protein contain the same information expressed in different ways.<\/figcaption><\/figure>\n\n\n<div class=\"wp-block-image\">\n<figure class=\"aligncenter size-full\"><img decoding=\"async\" width=\"567\" height=\"276\" src=\"https:\/\/adntro.com\/wp-content\/uploads\/2022\/04\/metodologia-vacunas.png\" alt=\"Vaccine methodology\" class=\"wp-image-6252\" srcset=\"https:\/\/adntro.com\/wp-content\/uploads\/2022\/04\/metodologia-vacunas.png 567w, https:\/\/adntro.com\/wp-content\/uploads\/2022\/04\/metodologia-vacunas-300x146.png 300w, https:\/\/adntro.com\/wp-content\/uploads\/2022\/04\/metodologia-vacunas-18x9.png 18w\" sizes=\"(max-width: 567px) 100vw, 567px\" \/><figcaption class=\"wp-element-caption\">Source: WHO<\/figcaption><\/figure>\n<\/div>\n\n\n<p>There are three main methods of making a vaccine:<\/p>\n\n\n\n<h2 class=\"wp-block-heading\">Attenuated vaccines<\/h2>\n\n\n\n<p class=\" translation-block\"><p><span style=\"font-weight: 400\">In this type of vaccine, the <\/span><strong>whole agent is used<\/strong><span style=\"font-weight: 400\">. It would be the method used to develop the smallpox vaccine, but more sophisticated. For this method there are different approaches generating different types of vaccines:<\/span><\/p><\/p>\n\n\n\n<ol class=\"wp-block-list\">\n<li class=\" translation-block\"><p><strong>Inactivated vaccines<\/strong><span style=\"font-weight: 400\">: consists of inactivating or destroying the pathogenic virus by chemical substances, heat or radiation. An example of a vaccine developed with this methodology is that of influenza.<\/span><\/p><\/li>\n\n\n\n<li class=\" translation-block\">Attenuated vaccines: the pathogen remains weakened but active causing a much smaller infection. Sometimes this type of vaccine is not recommended for immunocompromised people and among this type of vaccines we find chickenpox.<\/li>\n<\/ol>\n\n\n\n<h2 class=\"wp-block-heading\">Viral vector vaccines<\/h2>\n\n\n\n<p class=\" translation-block\"><p><span style=\"font-weight: 400\">In this case <\/span><strong>a fragment of the introduced pathogen<\/strong><span style=\"font-weight: 400\"> is used with harmless viral vectors, which are used to transport a specific fragment of the pathogen's DNA. The<\/span><strong> harmless virus<\/strong><span style=\"font-weight: 400\"> serves <\/span><strong>as transport<\/strong><span style=\"font-weight: 400\"> (vector) to introduce the pathogenic virus protein into the body. The Ebola vaccine is based on this methodology. <\/span><\/p><\/p>\n\n\n\n<h2 class=\"wp-block-heading\">mRNA vaccines<\/h2>\n\n\n\n<p class=\" translation-block\"><p><span style=\"font-weight: 400\">It uses the genetic material of the virus, the <\/span><strong>instructions for making virus-specific proteins<\/strong><span style=\"font-weight: 400\">. In this type of vaccine, both DNA and mRNA can be used. The mRNA is something temporary that is destroyed in a matter of hours (low stability), so it remains in our body only the time necessary to express the protein against which we are going to generate immunity.<\/span><\/p><\/p>\n\n\n\n<h2 class=\"wp-block-heading\">Vaccines against COVID-19<\/h2>\n\n\n\n<p>Given the pandemic situation that has been with us for more than two years, different vaccines have been developed using 2 of the previous methodologies: mRNA and viral vectors.<\/p>\n\n\n\n<p>Currently 3 vaccines stand out worldwide (Pfizer, Moderna and J &amp; J).<\/p>\n\n\n\n<p class=\"has-medium-font-size\"><strong>J&amp;J: Viral vector vaccine<\/strong><\/p>\n\n\n\n<p>The virus that is used to carry the SARS-CoV-2 virus fragment is a <strong>adenoviruses<\/strong> similar to the virus that causes the common cold, but to which <strong>important parts of the viral genome have been removed so that it cannot replicate in our body, so that it cannot infect <\/strong>and therefore cannot cause the common cold.<\/p>\n\n\n\n<p>The <strong>fragment that we incorporated into the adenovirus<\/strong> is what is really specific to each vaccine. Our cellular machinery will read that mRNA, that is, those instructions to generate the spike protein against which our body will produce antibodies.<\/p>\n\n\n\n<p>If we remember the central dogma of molecular biology, mRNA is the form of the genetic content that gives rise to proteins. However, there is <\/span><strong>great interindividual variability<\/strong><span style=\"font-weight: 400\"> when we talk about the reaction suffered after being<\/span><strong> vaccinated against COVID-19<\/strong><span style=\"font-weight: 400\">.<\/span><\/p><\/p>\n\n\n\n<p>A scientific study has been carried out to elucidate which genetic variants are associated with this variability. The three main vaccines (Pfizer, Moderna and J&amp;J) have been analyzed. However, because the sample size for J&amp;J was too low in this study, it was excluded from the analysis.<\/p>\n\n\n\n<p class=\" translation-block\"><p><span style=\"font-weight: 400\">The scientific study reveals that <\/span><strong>HLA-A*03:01 <\/strong><span style=\"font-weight: 400\">(HLA: encodes the cell surface proteins responsible for the regulation of the immune system) is the <\/span><strong>strongest genetic association<\/strong><span style=\"font-weight: 400\"> for people who received the <\/span><strong>Pfizer<\/strong><span style=\"font-weight: 400\"> vaccine and suffered<\/span><strong> extreme difficulties in carrying out their daily routine after vaccination against COVID-19<\/strong><span style=\"font-weight: 400\">. All\u00ed estudi\u00f3 las redes de interacci\u00f3n metab\u00f3lica en el microbioma de pacientes con Enfermedad de Chron y Colitis Ulcerosa en el centro de investigaci\u00f3n CBGP, centro de Excelencia Severo Ochoa (2020).
